These results could revolutionize approaches to preventing age-related cognitive decline.

The changes in appearance during aging occur gradually. However, when it comes to the brain, the process is less smooth and predictable. According to a recent study by an international team of scientists, there is a point at which it begins to age much faster before stabilizing again.

The team analyzed functional connectivity between brain regions in over 19,300 individuals across four large datasets. Their findings indicate that brain networks degrade according to a statistical S-shaped curve with clear transition points, rather than a clinical onset in late life or a gradual linear decline as previously assumed.

The effect first manifests around the age of 44, with degeneration reaching peak acceleration by 67 and plateauing by 90. Previous work by the team led by co-author Nathan Smith showed that this process is significantly influenced by energy loss in neurons (hypometabolism). Thus, the transition points suggest that there are specific windows when interventions may be most effective.

The team has already tested this in a clinical study with 101 participants of varying ages, who were given either ketones or glucose and underwent fMRI scans. Unlike glucose, ketones effectively stabilized deteriorating brain networks, but with effects that varied significantly at critical transition points. Ketones showed:

  • moderate benefits in young individuals (20-39 years);
  • maximum benefits during the "metabolic stress" of middle age (40-59 years), after which networks begin to destabilize;
  • reduced impact in older adults (60-79 years), just as network destabilization reached peak acceleration and complex vascular effects dominated.

Mujica-Parodi and co-authors stated that these results could revolutionize approaches to preventing age-related cognitive decline and neurodegenerative diseases such as Alzheimer's disease.

Current treatment methods typically target symptoms after they appear, often too late for meaningful intervention. This research suggests that metabolic intervention—whether through dietary approaches like ketogenic diets or supplements—may be most effective if initiated at age 40, long before cognitive symptoms arise.

Notably, in last year's study, researchers from Stanford University also identified age 44 as the first turning point in the aging of the body.
